Well, a couple things. 

First, you should post this on the Lister Engines forum.  This one is off in the sticks.  Nobody's going to see it here.  Go back to the home page, click on Lister Engine Forum, click on Listeroid Engines, and post there.  That's where all the creatures gather.

Second, you need to be more specific.  I guarantee you'll be asked the following:

Has the engine ever run before or are you setting it up for the first time?
What color is the smoke?  Choose one:  a. White, or  b.  Black
Have you checked the linkage to make sure the fuel rack on the dead cylinder is, in fact opening?
Have you swapped injectors to make sure it's not a bad injector?
Have you checked the injection timing on the dead cylinder?
Have you checked the valve clearances?
Are you sure you're getting compression?

...and a bunch more questions that I can't think of right at the moment.

Toss out some info where I told you and we'll have you thumpin' in no time.

In 90% of cases where a new twin is missing the cause is misadjusted throttle linkage. If one cylinder's exhaust is colder than the other undo the throttle clevis on the cold side and screw it in a half turn. Tighten the locknut and reassemble.  Keep doing so until both cylinders are the same temp.
