There's a screw and locknut below the injection pump which probably applies pressure to the pump tappet, what's it for?

I believe you are talking about the alignment guide for the tappet. The pump tappet is a roller-bearing type and not designed to rotate like the valve tappets... it must be held in proper alignment to the cam lobe.
